Banana Peel Oxalate Oxidase-detection, Purification, Characterization and Physiological Role

چکیده
Objective: Enzymes like oxalate oxidase (EC 1.2.3.4) and superoxide dismutase (EC 1.15.1.1) from germin family are known to generate active oxygen species. In the mammalian system, excess accumulations of oxalate causes kidney stones. Oxalate oxidase, an H2O2-generating enzyme, used for detection of oxalate. The aim of the present work is to screen out the activity of enzymes from all three stages of banana (Musa paradisica L. Variety “Bhusawal”) peel and to isolate, purified and characterized oxalate oxidase from this. With that describe the physiological role of both oxalate oxidase and superoxide dismutase in the plant. Methods: Oxalate oxidase activity can be detected directly in SDS-PAGE gel. Purification was done by using ion-exchange chromatography and SDSPAGE Gel. Results: Highest activity 5.99+0.021 unit /mg of oxalate oxidase were detected in leaky ripe stage of banana peel after purification. In crude extract of unripe banana peel activity of superoxide dismutase were found high (2.41unit/mg) compared to oxalate oxidase (0.269+ 0.020 unit/mg). Their occurence in different ripening stage of banana peel shows its role in plant defense mechanism. Conclusion: The purified enzyme of oxalate oxidase from banana peel is useful in the determination of oxalate content in common food, which is necessary for the prescription of the low oxalate diet for a patient with urinary and kidney stone where as superoxide dismutase work against ageing.

Five treatment procedures evaluated for the elimination of ascorbate interference in the enzymatic determination of urinary oxalate.
We evaluated the efficacies of five treatment procedures for eliminating ascorbate interference in the enzymatic determination of urinary oxalate. Aliquots of urine samples, containing different amounts of added ascorbate and oxalate, were individually subjected to ferric chloride, sodium nitrite, sodium periodate, charcoal, or ascorbate oxidase treatment to eliminate ascorbate interference. Ox...
متن کاملPurification and Characterization of Α-amylase Produced by Aspergillus Niger Using Banana Peels
The present study was aimed at purification and characterization of α-amylase enzyme from strain of Aspergillus niger isolated from local soil samples. Solid state fermentation (SSF) was carried out to produce α-amylase from A. niger using waste banana peel as substrate. The maximum activity of α-amylase (11926 U/gds) was recorded after 72 hours of fermentation. The extracted enzyme was subject...
متن کاملRemoval of Basic Blue 159 from Aqueous Solution Using Banana Peel as a Low-Cost Adsorbent
In this paper, the adsorption of Basic Blue159 (BB159) onto banana peel as a low-cost material was studied. At first, the banana peel was sieved. Later, banana peel particles were characterized by field emission scanning electron microscopy (FESEM), energy dispersive x-ray spectroscopy (EDXS) and Fourier Transform Infrared (FTIR) techniques. Batch adsorption experiments were carried out as a fu...
متن کاملExtraction and characterization of pectin from Saba banana [Musa ‘saba’(Musa acuminata x Musa balbisiana)] peel wastes: A preliminary study
Saba banana, Musa ‘saba’(Musa acuminata x Musa balbisiana) peel is an underutilized waste from processed banana manufacturing in the Philippines. The feasibility of saba banana peel pectin for use in food processing was assessed. Pectin from saba banana peel was extracted under two different extraction conditions. Pectin extraction was carried out using hydrochloric acid (0.5N, pH 1.5) and citr...
متن کاملGlobal Transcriptomic Analysis of Targeted Silencing of Two Paralogous ACC Oxidase Genes in Banana
Among 18 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase homologous genes existing in the banana genome there are two genes, Mh-ACO1 and Mh-ACO2, that participate in banana fruit ripening. To better understand the physiological functions of Mh-ACO1 and Mh-ACO2, two hairpin-type siRNA expression vectors targeting both the Mh-ACO1 and Mh-ACO2 were constructed and incorporated into the banana ...
